ABSTRACT

Aim To study the effect of emodin in Po-lygonum multiflorum on the expression of CYP450 isoenzymes in L02 hepatocytes and explore its mecha-nism of cytotoxicity. Methods L02 cells were treated with different concentrations of emodin. Cell viability was examined by MTS assay kit, and cell membrane injury was examined by detecting the release rate of lactate dehydrogenase( LDH) . The expression of cyto-chrome P450 mRNA was detected by real time PCR. Results The result of MTS assay showed that L02 cells viability was significantly reduced following expo-sure to emodin in a concentration and time dependent manner. The LDH release rate of L02 cells significant-ly increased after exposure to emodin for 48 h com-pared with the control group. On the mRNA level, compared with the control group,emodin had inductive effects on mRNA of each CYP450 enzyme, while had significant inductive effects on mRNA of CYP1 A1 and CYP1 B1 in a concentration and time dependent man-ner. Conclusion Emodin in Polygonum multiflorum may generate significant liver injury in L02 cells and has inductive effects on CYP450 enzyme activity.

ABSTRACT

Kawasaki disease (KD) is an acute systemic vasculitis that primarily affects young children aged from 6 months to 4 years.KD may lead to coronary artery abnormalities in 25%-30% of the children.The timely use of high dose intravenous immunoglobulin (IVIG) can significantly reduce the incidence of coronary artery aneurysms.Most clinicians use IVIG combined with aspirin as conventional therapy.But how to deal with those IVIG-resistant patients is a dilemma.With the deepening of research,many researchers have found that some second-line or third-line drugs such as corticosteroids,tumor necrosis factor-α inhibitors,and calcineurin inhibitors work better,and can effectively reduce the complication rate.The recent advances of the treatment of KD were summarized in this paper.

ABSTRACT

Kawasaki disease (KD) is an acute systemic vasculitis that primarily affects young children between 6 months and 4 years old. Coronary arteritis is an important clinical feature of KD because it is associated with aneurysms and thromboembolic events that can lead to severe complications, even sudden death. To date, the etiology and pathogenesis of Kawasaki disease has not been understood completely. In this paper, we will review the recent advances in epidemiology, etiology, pathogenesis and genetic susceptibility of Kawasa-ki disease.

ABSTRACT

A catheter(0.3 mm O.D.)was introduced into the external carotid artery(ECA) lumen into the lumen of the internal carotid artery(ICA), then it was gently advanced to the intracranial segment of ICA until the intraluminal catheter was 2~3mm from the origin of MCA. At this point thrombin were injected into ICA to induce thrombosis. Behavior disorders were observed at 3, 6, 12h after the operation. Willis circle, infarct volume and histopathologic changes were examined 12 hours after operation. The results showed that the model had stable behavior disorder score and infarct volume. Mixed thrombus could be observed at the origin of the MCA. Infarction volume of this model was similar with that of the model developed by linear occlusion of MCA in our laboratory. The model has the advantages of easy operation, reliable ischemia and reproducible infarct volume. It may be useful in documenting the safety and efficacy of thrombolytic therapies.
